In the course of studying the incidence of k-ras mutations in DNA in the circulation of patients with colorectal disease, we observed that our ability to detect mutated k-ras DNA in serum varied with the method of DNA isolation. Two methods of DNA isolation were used in this study. One was the QIAamp DNA Blood Midi Kit (Qiagen, Inc.), which has been widely used for isolating circulating DNA from serum or plasma (1–5). The other is the modified Guanidine/Promega Resin (G/R) method that was developed to isolate DNA from urine (6). To compare the applicability of the Qiagen and G/R methods for isolation of circulating DNA, we purified DNA from aliquots of serum from six patients with known mutant k-ras-positive colorectal disease (colorectal cancer or adenomatous polyps) and analyzed it for the presence of mutated k-ras …

Detection of mutated K-ras DNA in urine, plasma, and serum of patients with colorectal carcinoma or adenomatous polyps.
Our previous studies demonstrated that urine contains DNA derived from the circulation and that this DNA originated, in part, from organ sites and tumors distal to the urinary tract. To explore the potential use of DNA from urine as compared to other body fluids as a source for circulating DNA for cancer detection, the DNA concentration and the frequency of detection of mutated Kristin-ras (K-r...
متن کاملSignificance of Liquid Biopsy for Monitoring and Therapy Decision of Colorectal Cancer
PURPOSE Despite therapeutic improvements, all patients with nonresectable metastatic colorectal cancer (mCRC) acquire resistance to treatment probably due to the growth of mutated clones. In contrast to tissue-based studies, liquid biopsies have enabled the opportunity to reveal emerging resistance to treatment by detecting mutated clones and noninvasively monitoring clonal dynamics during ther...
متن کاملDietary phytate lowers K-ras mutational frequency, decreases DNA-adduct and hydroxyl radical formation in azoxymethane-induced colon cancer
Objective(s): Dietary phytate is known to protect against azoxymethane (AOM)-induced preneoplastic lesions. The present study was designed to determine whether dietary phytate affects mutation frequency in colon epithelial cells challenged with azoxymethane in vivo, through lowering the formation of O6-methyl guanosine (O6-MeG) and 8-hydroxy deoxyguanosine (8-OHdG) ad...
متن کاملRemoval of high-molecular-weight DNA by carboxylated magnetic beads enhances the detection of mutated K-ras DNA in urine.
We have previously demonstrated that mutated DNA derived from the circulation can be detected in urine and predominantly exists as DNA fragments <1 kb. To preferentially isolate the trans-renal DNA from urine, we developed a method using carboxylated beads to separate high-MW (1 kb or larger) from low-MW DNA in urine. A primer set for 18s rRNA (generating a PCR product of 872 bp) was designed a...
متن کاملMutations in K-ras codon 12 detected in plasma DNA are not an indicator of disease in patients with non-small cell lung cancer.
To the Editor: The demonstration that cell-free circulating DNA detected in the plasma of cancer patients is genetically identical to that of the primary tumor has generated substantial interest, leading to 200 publications (http:// www.ncbi.nlm.nih.gov). Recently, Wang et al. (1 ) reported in this journal that the method chosen for DNA isolation might contribute significantly to mutation detec...
